Dark chocolate lovers can handle a wider range of bitter tastes before rejection compared to milk chocolate fans, according to Penn State food scientists.

In a test of bitterness rejection levels in chocolate, people who prefer milk chocolate quickly detected -- and disliked -- milk chocolate with a bitter substance added to the candy, according to Meriel Harwood, a graduate student in food science. Dark chocolate fans had significantly higher tolerance to the added bitterness than people who like milk chocolate.

"In some cases, you may be able to detect a change in the taste of your food, but that might not necessarily lead to disliking the product," said Harwood. "However, almost immediately, people who preferred milk chocolate indicated they tasted something different and they didn't like it."

The researchers, who report their findings in the online version of the Journal of Food Science, divided a group of 85 participants into two groups based on their self-identified preferences in chocolate. A total of 43 people told researchers they preferred milk chocolate, and 42 people said they preferred dark chocolate.

Participants tried a series of pairs of dime-sized chocolate samples. In each pair, one sample contained sucrose octaacetate -- SOA -- a bitter-tasting substance, and the other did not. In each successive pair, the next samples contained increasing amounts of SOA.

The milk chocolate group quickly rejected the samples with SOA, while the dark chocolate group continued to like the candy, Harwood said. The dark chocolate group had a rejection threshold more than 2.5 times the rejection threshold level of those who prefer milk chocolate.

Harwood, who worked with Greg Ziegler, professor of food science, and John Hayes, assistant professor of food science and director of the sensory evaluation center, said that tests of rejection thresholds in food may be a simpler and more direct way to test food acceptability rather than measuring when the consumer can taste something different -- detection thresholds. The detection threshold test is one of the most common taste tests used in sensory science and is usually paired with affective -- or, consumer -- testing to determine preferences.

People do not always dislike a food because they detect something different, even if that difference may be considered a defect or undesirable attribute, according to Harwood.

"There may be a disconnect between preferences and the ability to detect tastes, like bitterness," said Harwood. "In other words, using detection threshold tests may not predict consumer acceptability."

Previous studies on cork taint in wine have shown that even though wine drinkers tasted the cork taint, they did not necessarily find the wine objectionable.

"We bring a bunch of cultural and emotional baggage with us when we eat," said Harwood. "What you grow up with, what you are used to, what you know, for example, can all influence your preferences."

Harwood said rejection threshold tests may lead to cost-savings for the food industry.

Rather than disposing products that are thought to be undesirable and wasting costly materials, researchers could use rejection thresholds to find out whether consumers will object to the taste of a product. Food manufacturers may also be able to use alternative ingredients, such as sugar or salt replacements, if they can first identify a rejection threshold level for those ingredients in products. This could give product developers more versatility when they create recipes.

He says that when disaster strikes, "we're not Democrats or Republicans first, we are Americans first."

The president says during a campaign event in Ames, Iowa, that preparations for the storm have been under way for days, and he vows the nation will "help our neighbors in need."

Obama has warned residents of the Gulf Coast to listen to local authorities and follow their directions ahead of the hurricane that is veering toward New Orleans.

Gambia on edge amid mass executions

Gambia's President Yahya Jammeh came under attack Tuesday for sending nine prisoners to the firing squad, leaving the tiny nation jittery as another 38 convicts await execution in the coming weeks.

The 47-year-old former soldier, who seized power in a 1994 coup, dismissed concerns expressed by the African Union and rights bodies that had urged him not to carry out his plans and executed a first batch of prisoners on Sunday night.

"We are absolutely dumbfounded by this news," said Alioune Tine of the Dakar-based African Assembly for the Defense of Human Rights.

"It is a case of madness at the head of a country, a paranoiac at the head of Gambia and that is dangerous for Africa," Tine said.

Calling for the "total isolation" of the Gambian leader, who often heaps derision on criticism from the West, Tine urged a halt to meetings of the African Commission on Human and People's Rights, which has its headquarters in Banjul.

Gambia's interior ministry announced Monday that nine death row prisoners had been executed by firing squad on Sunday night, a week after Jammeh vowed to carry out all death sentences by mid-September.

Two of those killed, one of them a woman, were Senegalese citizens, and Senegal's President Macky Sall said he "deeply regrets" the killings, in a statement from his spokesman.

"Macky Sall calls on Gambia to urgently suspend all executions," read the statement.

Another Senegalese citizen is still on death row in Gambia, which is surrounded by Senegal except for a strip of Atlantic coast.

Commonwealth spokesman Richard Uku said in a statement the executions were "a matter of grave concern" and urged the government of the former British colony, a member of the 54-state Commonwealth, to renounce its plans.

The European Union has also demanded "an immediate halt" to the executions.

Amnesty International estimates a further 38 prisoners are on death row.

"We are appalled that the Gambian authorities carried out the nine executions and urge them to ensure that no further executions take place," said Paule Rigaud, the rights group's deputy director for Africa.

Jammeh, who has woven an aura of mysticism around himself, dressing in billowing white robes and always clutching his Koran, is accused of ruling the tiniest nation on the African mainland with an iron fist, brooking no dissent.

The president's feared plain-clothes security officers could be seen on the streets of Banjul on Tuesday, and tension was palpable among citizens, who were afraid to talk about the executions lest they be recorded by his agents and punished.

In a televised address to mark this year's Muslim feast of Eid al-Fitr, Jammeh said: "By the middle of next month, all the death sentences will have been carried out to the letter.

"There is no way my government will allow 99 percent of the population to be held to ransom by criminals."

The regime of the man who says he can cure AIDS is often pilloried for human rights abuses, enforced disappearances, extra-judicial killings, torture and muzzling journalists.

Many top officials have found themselves charged with treason, often related to coup plots which observers have said are a sign of paranoia by Jammeh, who won a fourth term in office in November 2011.

Last year eight military top brass, including the former army and intelligence chiefs and the ex-deputy head of the police force, were sentenced to death for treason.

Jammeh also regularly reshuffles top officials.

On Sunday night state television reported that Education Minister Mambury Nie had been sacked a week after being moved into the portfolio from the foreign ministry.

In the past few months former army chiefs, heads of the navy and presidential guard and other generals have been sacked and deployed as deputy ambassadors around the world.

Prior to Sunday, the last official execution in Gambia was in 1985 and Amnesty International had considered Gambia to be among the 22 of Africa's 54 states that had in practice abolished capital punishment.

The nation of 1.7 million people survives mostly off tourism, luring sun-worshipping Europeans to its sweeping, palm-fringed coastline, and agriculture.

Sleep learning is possible: Associations formed when asleep remained intact when awake

ScienceDaily (Aug. 26, 2012) ? Is sleep learning possible? A new Weizmann Institute study appearing August 26 in Nature Neuroscience has found that if certain odors are presented after tones during sleep, people will start sniffing when they hear the tones alone -- even when no odor is present -- both during sleep and, later, when awake. In other words, people can learn new information while they sleep, and this can unconsciously modify their waking behavior.

Sleep-learning experiments are notoriously difficult to conduct. For one thing, one must be sure that the subjects are actually asleep and stay that way during the "lessons." The most rigorous trials of verbal sleep learning have failed to show any new knowledge taking root. While more and more research has demonstrated the importance of sleep for learning and memory consolidation, none had managed to show actual learning of new information taking place in an adult brain during sleep.

Prof. Noam Sobel and research student Anat Arzi, together with Sobel's group in the Institute's Neurobiology Department in collaboration with researchers from Loewenstein Hospital and the Academic College of Tel Aviv -- Jaffa, chose to experiment with a type of conditioning that involves exposing subjects to a tone followed by an odor, so that they soon exhibit a similar response to the tone as they would to the odor. The pairing of tones and odors presented several advantages. Neither wakes the sleeper (in fact, certain odors can promote sound sleep), yet the brain processes them and even reacts during slumber. Moreover, the sense of smell holds a unique non-verbal measure that can be observed -- namely sniffing. The researchers found that, in the case of smelling, the sleeping brain acts much as it does when awake: We inhale deeply when we smell a pleasant aroma but stop our inhalation short when assaulted by a bad smell. This variation in sniffing could be recorded whether the subjects were asleep or awake. Finally, this type of conditioning, while it may appear to be quite simple, is associated with some higher brain areas -- including the hippocampus, which is involved in memory formation.

In the experiments, the subjects slept in a special lab while their sleep state was continuously monitored. (Waking up during the conditioning -- even for a moment -- disqualified the results.) As they slept, a tone was played, followed by an odor -- either pleasant or unpleasant. Then another tone was played, followed by an odor at the opposite end of the pleasantness scale. Over the course of the night, the associations were partially reinforced, so that the subject was exposed to just the tones as well. The sleeping volunteers reacted to the tones alone as if the associated odor were still present -- by either sniffing deeply or taking shallow breaths.

The next day, the now awake subjects again heard the tones alone -- with no accompanying odor. Although they had no conscious recollection of listening to them during the night, their breathing patterns told a different story. When exposed to tones that had been paired with pleasant odors, they sniffed deeply, while the second tones -- those associated with bad smells -- provoked short, shallow sniffs.

The team then asked whether this type of learning is tied to a particular phase of sleep. In a second experiment, they divided the sleep cycles into rapid eye movement (REM) and non-REM sleep, and then induced the conditioning during only one phase or the other. Surprisingly, they found that the learned response was more pronounced during the REM phase, but the transfer of the association from sleep to waking was evident only when learning took place during the non-REM phase. Sobel and Arzi suggest that during REM sleep we may be more open to influence from the stimuli in our surroundings, but so-called "dream amnesia" -- which makes us forget most of our dreams -- may operate on any conditioning occurring in that stage of sleep. In contrast, non-REM sleep is the phase that is important for memory consolidation, so it might also play a role in this form of sleep-learning.

Although Sobel's lab studies the sense of smell, Arzi intends to continue investigating brain processing in altered states of consciousness such as sleep and coma. "Now that we know that some kind of sleep learning is possible," says Arzi, "we want to find where the limits lie -- what information can be learned during sleep and what information cannot."

There are many ways to create three dimensional objects on a micrometer scale. But how can the chemical properties of a material be tuned at micrometer precision? Scientists at the Vienna University of Technology developed a method to attach molecules at exactly the right place. When biological tissue is grown, this method can allow the positioning of chemical signals, telling living cells where to attach. The new technique also holds promise for sensor technology: A tiny three dimensional "lab on a chip" could be created, in which accurately positioned molecules react with substances from the environment.

Materials Science and Chemistry

"3D-photografting" is the name of the new method. Two research teams from the Vienna University of Technology collaborated closely to develop it: Professor Jrgen Stampfl's materials science team and Professor Robert Liska's research group for macromolecular chemistry.

Both research groups have already attracted considerable attention in the past, developing new kinds of 3D-printers. However, for the applications on which the scientists are working on now, 3D-printing would not have been useful: "Putting together a material from tiny building blocks with different chemical properties would be extremely complicated", says Aleksandr Ovsianikov. "That is why we start from a three dimensional scaffold and then attach the desired molecules at exactly the right positions."

Molecules in the Hydrogel Locked into Position by the Laser

The scientists start with a so-called hydrogel a material made of macromolecules, arranged in a loose meshwork. Between those molecules, large pores remain, through which other molecules or even cells can migrate.

Specially selected molecules are introduced into the hydrogel meshwork, then certain points are irradiated with a laser beam. At the positions where the focused laser beam is most intense, a photochemically labile bond is broken. That way, highly reactive intermediates are created which locally attach to the hydrogel very quickly. The precision depends on the laser's lens system, at the Vienna University of Technology a resolution of 4 m could be obtained. "Much like an artist, placing colors at certain points of the canvas, we can place molecules in the hydrogel but in three dimensions and with high precision", says Aleksandr Ovsianikov.

Chemical Signals for Cells

This method can be used to artificially grow biological tissue. Like a climbing plant clinging to a rack, cells need some scaffold at which they attach. In a natural tissue, the extracellular matrix does the trick by using specific amino acid sequences to signal the cells, where they are supposed to grow.

In the lab, scientists are trying to use similar chemical signals. In various experiments, cell attachment could be guided on two dimensional surfaces, but in order to grow larger tissues with a specific inner structure (such as capillaries), a truly three dimensional technique is required.

Micro Sensors Detect Molecules

Depending on the application, different molecules can be used. 3D photografting is not only useful for bio-engineering but also for other fields, such as photovoltaics or sensor technology.
